For me Anything in the first episodes of Attack on Titan, since so many important characters die, which continues into the next seasons with less effective results The ending to Devilman Crybaby (though those who are familiar with the original manga will know what happens already) Gon vs. Pitou in Hunter X Hunter for being the most grim and gruesome beating in the show ever Deku vs. Muscular in My Hero Academia, as well as Ochako vs. Bakugo in that anime's tournament arc The identity of Dandy in Space Dandy for subverting expectations and explaining all the art shifts and retcons in a way that works anything in Land of the Lustrous for convincing me for the first time in my life that CGI can be beautiful in anime

I must be getting old because I watched the pilot without knowing anything about the characters going in, and didn't at all get that Charlie and Vaggie are in a lesbian relationship. I suppose the "I don't touch the gays" line should've tipped me off, but I must've missed that on my first viewing because every single beat has the exact same pacing.

You may be onto something. This story has been existing online for so long, people watching the pilot have been familiar with even the EXTRAS who only appear only once!
